NS5 Protein

NS5, the largest non-structural protein at 103 kDa, consists of an N-terminal methyltransferase (MTase) domain with three activities essential for cap synthesis (guanylyltransferase, guanine-N7-methyltransferase, and nucleoside-2’O-methyltransferase) and a C-terminal RNA-dependent RNA polymerase (RdRp) domain responsible for de novo RNA synthesis. The NS5 protein displays high conservation across the four virus serotypes, and due to the absence of human enzymes with RdRp activity, NS5 has been identified as a suitable target for anti-dengue drug development.
